A. Personal Statement

    I have over 10 years of expertise in and contribute to the understanding of molecular and biophysical mechanisms of ion channel activation and the translational research of disease-associated ion channel. Two ion channel families, TRP channels and BK type voltage and Ca2+ activated K+ channels have been the model systems for my studies. My research has been published in international top-tier, peer-reviewed journals including PNAS, JGP, J Neurosci, Oncology and others. One of my pioneer research is highly cited (51 cites over world), and was cited as critical proof by FDA (2013).
    In the translational studies of TRP channels, I first observe TRPM8 ion channel functionally expressed in bladder cancer, and its natural agonist, menthol, can induce the programmed death (apoptosis) of bladder cancer T24 cells directly via TRPM8 channel (Li Q et al. 2009, Oncology,77(6):335-341). And in my early postdoc program, we firstly discovered that TRPM7 and TRPM8 expression are significantly correlated with the clinical characteristics of pancreatic tumor and the malignant characteristics of pancreatic cancer(Yee et al. 2014,Cells, 3(2): 500–516; 2015, Biol Open 15; 4(4): 507–514). Recently, we found that TRPV1 /TLR4 pathway play a critical role in sensory neuron responses to Paclitaxel (Li et al. 2015,J Neurosci, 30;35(39):13487-500).
    In the study of BK channels, we systematically elucidate the molecular regulatory mechanisms of BK γ-subunits on BK channel. We have identified the critical structural and functional mechanisms of γ1-subunit to modulate BK channels (Li et al. 2015, J Gen Physiol,145(6): 543–554; 2016, J Gen Physiol,147 (4):337-351). And we also figure out the interaction part of BKαto interact with γ1-subunit (not published), and the role of the extracellular LRR loop to modulate BK channel’s activities (not published) as well. More interesting, we recently found BK channels can be conditionally induced into C-type inactivation, which provide the molecular structure to understand BK channel (Yan et al. 2016, PNAS,113(25):6991-6996).
